目的 探究岩盐气溶胶对肺炎幼鼠炎症反应和肺损伤的治疗作用,并初步探究其机制。方法 将30只BALB/c幼鼠随机分为对照组(Control组)、肺炎模型组(Model组)和岩盐气溶胶治疗组(Treat组),每组10只,通过鼻腔内滴注肺炎链球菌建立幼鼠肺炎模型;称量幼鼠体质量和肺湿质量并计算肺指数;HE染色和Masson染色观察幼鼠肺组织病理形态变化和纤维化情况;ELISA检测幼鼠BALF中TNF-α、IL-1β和sST2水平;RT-qPCR检测幼鼠肺组织miR-506-3p表达。结果 岩盐气溶胶治疗降低肺炎幼鼠肺指数,改善幼鼠肺组织病理损伤,减少幼鼠肺组织胶原纤维沉积,降低幼鼠BALF中TNF-α、IL-1β和sST2水平,增加幼鼠组织中miR-506-3p表达。结论 岩盐气溶胶治疗抑制肺炎幼鼠炎症反应和肺损伤,其机制与增加肺组织中miR-506-3p表达并抑制BLAF中sST2水平有关。
Abstract:Objective To explore the therapeutic effect of rock salt aerosols on inflammatory reaction and lung injury in mice with pneumonia, and to explore its mechanism preliminarily.Methods Thirty BALB/c mice were randomly divided into control group, pneumonia model group(Model group)and rock salt aerosols treatment group(Treat group),with 10 rats in each group.The model of pneumonia in mice was established by intranasal instillation of streptococcus pneumoniae.The body mass and wet lung mass of mice were weighed and the lung index was calculated.HE staining and Masson staining were used to observe the pathological changes and fibrosis of lung tissue in mice.ELISA was used to detect the levels of TNF-α,IL-1β and sST2 in BALF of mice.RT-qPCR was used to detect the expression of miR-506-3p in lung tissue of mice.Results Rock salt aerosols treatment could reduce lung index, improve pathological damage of lung tissue, reduce collagen fiber deposition in lung tissue of mice, reduce the levels of TNF-α,IL-1β and sST2 in BALF of mice, and increase the expression of miR-506-3p in mice.Conclusion Rock salt aerosols therapy inhibits inflammatory reaction and lung injury in mice with pneumonia, and its mechanism is related to increasing the expression of miR-506-3p in lung tissue and inhibiting the level of sST2 in BLAF.
